Synthesizes a gene of interest directly from sequence information without a template, with optional cloning into a specified vector. Supports synthesis of long fragments, high-GC, and repetitive sequences, with free codon optimization to improve target gene expression.
When the sequence you need is hard to obtain from an existing plasmid (for example, it requires codon optimization, is an artificially designed sequence, or the original source is unavailable), de novo gene synthesis lets you "build" that DNA directly to your design — answering "if nothing off-the-shelf exists, how do you get this sequence from scratch."
The target sequence is split into multiple overlapping oligonucleotides, assembled into a complete gene fragment by PCR or ligation-based assembly (Gibson Assembly/OE-PCR), then cloned into a vector and verified by full-length sequencing. Codon optimization uses the host's codon usage bias to adjust codon usage — without changing the amino acid sequence — removing rare codons and unfavorable secondary structures.
Protein expression, antibody genes, enzyme engineering, metabolic pathway construction, vaccine antigen design, etc.
Adapted for hosts such as E. coli, yeast, mammalian, and insect cells
