Verifies interactions between endogenous or exogenous proteins under near-physiological conditions; capable of detecting endogenous protein complexes while reasonably preserving physiological interaction relationships.
When yeast two-hybrid or bioinformatic prediction has suggested a possible interaction between two proteins, and you need to confirm it in an environment closer to real cellular conditions, Co-IP is the most commonly used verification method — answering "do these two proteins actually bind to each other inside the cell."
A specific antibody against the target protein (bait) is coupled to protein A/G magnetic beads or agarose to enrich the target protein and its bound interacting proteins (prey) from a cell lysate. The co-precipitated interacting proteins are then identified by Western blot or mass spectrometry, confirming the binding relationship between the proteins.
