Yeast One-Hybrid
Protein-DNA Interaction · Targeted Screening

We offer a targeted yeast one-hybrid verification service for studying the binding relationship between a protein (such as a transcription factor) and a specific DNA cis-element — a commonly used tool for screening protein-DNA interactions in transcriptional regulation research.

Purpose & Application Scenarios

Purpose

When you want to know exactly which proteins can bind a given DNA sequence — rather than verifying, in the other direction, the relationship between an already-known protein and a known DNA sequence — yeast one-hybrid helps you screen candidate binding proteins from a protein library — answering "who recognizes this DNA sequence."

Application Scenarios

Technical Principle & Applications

Technical Principle

A candidate DNA cis-element (bait) is cloned upstream of a reporter gene (such as HIS3 or LacZ) to construct a reporter vector, which is transformed into a yeast strain carrying a prey protein (such as a transcription factor) expression plasmid. If the prey protein binds the DNA element, it recruits the transcription machinery to activate reporter gene expression, and binding is judged by auxotrophic selection or a colorimetric reaction.

Applications

Transcription factor binding site verification, promoter/enhancer cis-element functional research, transcriptional regulatory network analysis, etc.

Service Advantages

Dedicated to Protein-DNA Interactions

Targeted at transcription factor-cis-element binding verification

Typically 2-4 Weeks

Covers vector construction, transformation, and screening

Reporter Gene Colorimetric Readout

Confirmed by auxotrophic selection or a colorimetric reaction

Connects to Downstream Verification

Positive results can be further confirmed with EMSA/ChIP

Delivery
