Candidate interacting proteins are each fused to the N- or C-terminal fragment of a fluorescent protein; if the two proteins interact, the fragments refold and restore fluorescence, allowing direct visualization of the protein interaction and its subcellular location in live cells.
Once Co-IP or another method has given you preliminary evidence that two proteins interact, and you want to directly "see" where and how that interaction occurs in a live cell, BiFC provides visual evidence — answering "where in the cell does this protein interaction actually happen."
A fluorescent protein (e.g., Venus/YFP) is split into non-fluorescent N- and C-terminal fragments, each fused to a candidate interacting protein. If the two proteins interact, the fragments are brought into proximity and refold into a functional fluorophore, producing a detectable fluorescence signal that directly reflects where and how strongly the interaction occurs.
